Fructose 1,6-bisphosphate is the two-phosphate sugar made by the committed step of glycolysis; aldolase splits it in two.

## Key facts

- Phosphofructokinase-1 makes it (step 3) using ATP; aldolase splits it into dihydroxyacetone phosphate and glyceraldehyde 3-phosphate.
- It activates pyruvate kinase ahead of time: what comes in at the top speeds the exit at the bottom.
- In gluconeogenesis, fructose-1,6-bisphosphatase turns it back into fructose 6-phosphate; fructose 2,6-bisphosphate and AMP inhibit it.

## Structure

- A furanose ring (five atoms) with phosphates on carbons 1 and 6; not to be confused with fructose 2,6-bisphosphate, which only regulates.

## Clinical notes

- Fructose-1,6-bisphosphatase deficiency causes fasting hypoglycemia and lactic acidosis, because it blocks gluconeogenesis.

## Test yourself

Think of the answer, then tap the question to see it.

## Which enzyme splits fructose 1,6-bisphosphate?

Aldolase, into dihydroxyacetone phosphate and glyceraldehyde 3-phosphate.

## Which enzyme does fructose 1,6-bisphosphate activate ahead of time?

Pyruvate kinase (step 10).

## How does it differ from fructose 2,6-bisphosphate?

The 1,6 form is a glycolysis intermediate; the 2,6 form is not metabolised: it only activates PFK-1 and inhibits fructose-1,6-bisphosphatase.

Molecules are shown in their neutral form, as the databases deposit them; in the cell the acids and phosphates are ionised.
